This study compared the pharmacokinetics, metabolism, and stress hormone responses of Hanford, Göttingen, Yucatan, and Sinclair minipigs after oral and IV dosing of a chiral test compound and found only minor (less than two-fold) differences in clearance, volume, bioavailability, and chiral inversion across strains. However, cortisol responses to a short-term stressor were lowest in the Sinclair minipig, suggesting that while PK differences are minimal, strain selection may be important depending on whether a study is sensitive to stress effects, particularly in chronic versus acute designs.
